Story:
As everyone knows, it's very difficult to characterize a Marvel character in more detail because of its existence in various Alternative Realities, therefore, without going into unnecessary details I will summarize the story of Coulson in his universe of origin, the MCU, also known as Earth-199999:
Phil Coulson was the right arm of Nick Fury, director of S.H.I.E.L.D., and had been with him in various situations, such as when appeared the Iron Man, Thor's hammer, and even in NY battle. In this battle (which, for those unaware, it passes in the first Avengers movie) Phil is seriously injured and killed by Loki.

His death motivates the Avengers, especially Captain America, which Phil was a big fan, and that's what keeps them together leading to victory against the Chitauri army.

Later, Coulson is resurrected and is entitled to his own team within S.H..I.E.L.D. With this team he spends lots of adventures until the fall of S.H.I.E.L.D. (events of Captain America: The Winter Soldier) where he is chosen by Fury himself, who everyone thought was dead, to be the new director.

From that moment on, he and his team, now reformulated, are charged wofsome Hydra threats and also of unravellingthe secret of the Inhumans ...

Phil was the first Marvel character to have origin in the films and then be converted to the comics (now having counterparts also in video-games and TV series, including animes), though now as a way to honor, some members his team have also been converted.

One last curiosity: in the main Marvel Universe (Earth-616) Phil has a diary with almost every known super heroe and their first appearance, and photos from newspapers and magazines, and articles to prove his data. Phil is so crazy about superheroes that one time he was taken prisoner, is found by his S.H.I.E.L.D. companions saying the name of everyone who once belonged to the X-Men in the chronological order of accession, scene that we'll hardly watch in live-action due to the fact that the rights of mutants (which includes the X-Men) belong to FOX.
